چکیده

assays for prostate cancer screening, Afssaps has conducted, in the context of its mission, a market survey of devices indicating quantitative results. The objective of this survey was to evaluate the analytical performances of these assays, particularly to verify the accuracy of total PSA, free PSA and complexed PSA assay kits and to study the equimolarity of total PSA reagents in the analogous recognition of the various forms of serum PSA (free PSA/bound PSA) by measuring 2 ng/ml, 4 ng/ml and 10 ng/ml samples of total PSA containing 0%, 25%, 50%, 75% or 100% of free PSA, prepared from Stanford University standard solutions (Stamey). Package leaflets were also examined to verify compliance with the essential requirements defined in Directive 98/79/EC.

Evaluation of a two-site immunoradiometric assay for measuring noncomplexed (free) prostate-specific antigen.

Serum prostate-specific antigen (PSA) in men is present as two different molecular forms separable by gel-filtration chromatography (GFC). We have evaluated a two-site IRMA that measures only the noncomplexed (free) form of PSA (F-PSA). Verification that the F-PSA assay measures solely F-PSA was obtained by assaying GFC-fractionated serum samples with both the F-PSA IRMA and a commercial PSA as...

Novel immunoassay for the measurement of complexed prostate-specific antigen in serum.

Serum prostate-specific antigen (PSA) is an effective diagnostic tool for detection of prostate cancer (CaP) at an early and potentially curable stage, but specificity is low. Studies have shown that the proportion of serum PSA complexed with alpha-1-antichymotrypsin (ACT) is higher in men with CaP than in men with benign prostate disease. We developed a novel immunoassay for complexed PSA base...

Dual-label one-step immunoassay for simultaneous measurement of free and total prostate-specific antigen concentrations and ratios in serum.

We developed a simple one-step dual-label immunoassay for simultaneous measurement of the free, noncomplexed form of prostate-specific antigen (PSA) and total PSA. The assay is based on time-resolved fluorescence and includes a stable fluorescent chelate of Eu to label a monoclonal antibody (mAb) that detects only free PSA, whereas a second mAb labeled with a fluorescent chelate of Tb provides ...

Epitope analysis of a prostate-specific antigen (PSA) C-terminal-specific monoclonal antibody and new aspects for the discrepancy between equimolar and skewed PSA assays.

BACKGROUND Immunoassays to measure prostate-specific antigen (PSA) often give different values for the same patient samples, and the calibrators among commercial immunoassays are not interchangeable. We developed three novel assays to quantify the free and complexed forms of PSA in serum. METHODS We synthesized 46 peptides, which encompassed the entire PSA molecule, and determined the interac...

Dual-label immunoassay for simultaneous measurement of prostate-specific antigen (PSA)-alpha1-antichymotrypsin complex together with free or total PSA.

BACKGROUND A major portion of prostate-specific antigen exists in circulation as a complex with alpha(1)-antichymotrypsin (PSA-ACT), whereas a minor part is free (fPSA). The proportion of PSA-ACT is increased in prostate cancer (PCa), but immunologic determination of PSA-ACT is hampered by a background produced by nonspecific adsorption of ACT to the solid phase. To reduce the nonspecific inter...
